Background information
Last week, we introduced application case of AT Protein A Diamond Ultra resin launched by Bestchrom.
Case study one: Efficient purification of bi-specific antibody
· Sample information: bi-specific antibody, molecular weight 148KDa
· Column: EzScreen 4.3mL, column bed height : 10cm
· Flow: 0.7 mL/min
· Buffer: PB and similar buffer, pH7.2
· Elution buffer: NaAc-HAc and similar buffer, pH 3.3
Dynamic binding capacity data
In this experiment, DBC of bi-specific antibody detection has been carried out on 12 types of resins available on the market. Get the DBC @ 10% break-through(see the following picture). As illustrated, DBC of AT Protein A Diamond Ultra can reach 43.88mg/mL, which is higher than that of resins from other brands. This enables larger sample volume being processed, boosting productivity.

Purification process and results
After comparing sample purity before/after purification, it is found that AT Protein A Diamond Ultra can effectively remove impurities and improving purity of target antibody. See the following figure for chromatogram and testing data.

Case study two: Efficient purification of monoclonal antibody
· Sample information: monoclonal antibody molecule, molecular weight 148KDa
· Column: EzScreen 4.3 mL, column bed height 10cm
· Flow: 0.7 mL/min
· Equilibration buffer: Tris-HCl and similar buffers, pH 7.2
· Cleaning buffer: citric acid and similar buffers, pH 5.5
Dynamic binding capacity data
In this experiment, monoclonal antibody was loaded on three types of affinity resins from Bestchrom. Get DBC @ 10% break-through data(see the following figure). As illustrated, Extrem A Diamond enjoyed the highest DBC(71.63mg/mL), while AT Protein A Diamond Ultra resin reached a DBC of 69.44mg/mL, a level close to Extrem A Diamond.

As illustrated by the application cases and data, AT Protein A Diamond Ultra enjoys exceptional performance in purification applications. From bench-scale lab experiment to process-scale purification project, the resin shows consistent performance stability and excellent purification productivity.
As application cases increase and feedbacks accumulate, AT Protein A Diamond Ultra will play a more vital role in bio-processing sector. The future will undoubtedly witness more technical break-through and innovation brought by Bestchrom to bio-pharmaceutical industry.
